Technology Use

The Cellexus single-use, disposable bioreactor technology which is applied in the Cellexus range of products has been uniquely designed to simplify, accelerate and improve cell culture and fermentation processes.

The Cellexus products include CellexusBags, which are single use, disposable cell culture bioreactor bags, and the CellMaker PLUS cell culture control instrumentation:

Use of the technology is very simple and, using the Cellexus CellMaker PLUS cell culture control instrumentation with an 8 litre Enclosure as an example, is briefly described below:

1. Load the CellexusBag into the CellMaker PLUS Enclosure

The CellexusBag is loaded into the CellMaker PLUS Enclosure. Access panels in the CellMaker PLUS Enclosure make it simple to insert the CellexusBag and locate the tubes and filters.

2. Lock the CellMaker PLUS Enclosure

The CellMaker PLUS Enclosure is closed and secured.

3. Fill the CellexusBag

For convenience a peristaltic pump is used to deliver pre-sterilised media to the CellexusBag or to deliver non-sterile media via a media filter (as shown).

4. Programme the temperature on CellMaker PLUS Controller

The touch screen of the CellMaker PLUS Controller can be programmed to set the appropriate temperature for the experiment. The CellMaker PLUS has the capacity to cool as well as heat the culture media (within the 15-40°C range).

5. Programme the CellMaker Oxygenator from the CellMaker PLUS Controller

The touch screen of the CellMaker PLUS Controller can be used to initiate the CellMaker Oxygenator. A purge cycle runs for 20 minutes before oxygen is available for delivery to the CellexusBag in the CellMaker PLUS Enclosure. The CellMaker Oxygenator is a key feature that enables optimal growth of oxygen-hungry bacteria and yeast. By including this as a standalone system, or dovetailed to the CellMaker PLUS SupplyHub, installation of the system is quick and simple without the need for, or additional cost of, external gas supply.

6. Adjust gas flows on the CellMaker PLUS Enclosure

Air and oxygen supplies fed from the CellMaker PLUS SupplyHub to the CellexusBag in the CellMaker PLUS Enclosure are controlled software on the CellMaker Controllor. Up to 10 litres/minute air and 4 litres/minute oxygen can be delivered to the CellexusBag via the sparge tube.

7. Inoculate the CellexusBag through the cell culture/sample ports on the CellMaker PLUS Enclosure

Culture/sample ports are included on the CellMaker PLUS Enclosure to facilitate easy inoculation and removal of samples from the cell culture in the CellexusBag.

8. Harvest cell culture through the media addition port on the CellMaker PLUS Enclosure

The CellexusBag cell culture bag will empty under gravity, internal gas pressure or via a peristaltic pump as required.
