Background & Overview
Cellexus provides an easy to use single-use, disposable bioreactor platform for cell culture and fermentation bioprocesses that can be used to accelerate and simplify cell culture and fermentation in biopharmaceutical research, scale up, production and manufacturing.
The single-use, disposable element of the Cellexus CellMaker PLUS product, and at the core of the Cellexus technology, is the CellexusBag which:
- Is a unique cell culture bioreactor bag designed for rapid set-up and optimisation of cell culture and fermentation processes under clean, sterile conditions without cross-contamination.
- Comes with sparge tube facility for efficient sample aeration and growth at high cell densities in E.Coli, pichia and algae. For culture of mammalian and insect cells, a modified CellexusBag with facility for pH and DO sensors can be used to maximise protein production efficiencies in the broadest range of cell types.
- Are made from a robust, co-extruded and multiple polymer material. They are manufactured from pharmaceutical grade plastics welded using a simple, strong manufacturing processes under class 10,000 clean room conditions and to the ISO standards required by the biopharmaceutical industry (Product File available for audit on request).
The CellexusBag has been constructed for use in the fully automated Cellexus CellMaker PLUS single-use, disposable bioreactor cell culture control instrumentation which includes four components:
- SupplyHub incorporating air pumping system, temperature regulation and data acquisition.
- Controller for important user defined parameters to be specified. The unit is modular and can be provided with / without DO and pH sensor reader options.
- Enclosure, in which the CellexusBag is held, which incorporates a Peltier heater to enable cell culture temperature to be closely monitored and controlled. Both 8L and 50L Enclosures are available to provide flexibility in scale.
- Oxygenator which dovetails with the SupplyHub to provide continuous supply of oxygen and which is also available as a stand-alone unit.
The Cellexus technologies employ a proprietary, novel airlift technology in the CellexusBag, which is simple, scalable and inexpensive to use. The main principles behind its use are:
- A patented air dispersion system on the CellMaker PLUS Enclosure aerates the cell culture from the base of the CellexusBag.
- The low density mixture of aqueous solution and gas forms a riser current and is heated via a heating plate located on the inner surface of the CellMaker PLUS Enclosure.
- Due to the unique shape of the CellexusBag, the upper surface acts as a deflector and pushes the riser current to the front of the CellexusBag upper chamber.
- The relatively large surface area of the CellexusBag, with respect to its volume, allows gentle gas exchange for cell cultures that do not require vigorous aerating.
- The low density riser current is displaced by the higher density down coming current.
- A temperature probe located on the CellMaker PLUS Enclosure on the opposite surface from the heating plate measures the temperature of the media. This data can be used to automatically regulate cell culture temperature from 15 to 40°C to within ±0.1°C.
